从拟南芥基因组中分别克隆AtCKX1基因和RD29B基因5′侧翼1705 bp启动子区域序列,生物信息学表明,AtCKX1含有黄素腺嘌呤二核苷酸(FAD)和细胞分裂素的结合位点;RD29B启动子片段中存在ABA响应元件(ABA response element;ABRE)、Myb结合位点、TATA盒、CAAT盒等顺式作用元件。分别将AtCKX1和RD29B插入载体pCAMBIA1390,构建了由RD29B驱动的AtCKX1的植物双元表达载体p1390RD29BAtCKX1。
Abstract:
In this study,the cloning of AtCKX1 and 5′flanking promoter sequence of droughtinducible RD29B from Arabidopsis was reported. The 1705 bp promoter sequence was analyzed bioinformatically in the database of plant cisacting regulatory element (PlantCARE). The result showed that there were several important cisacting elements,including ABRE (ABA response element),Myb binding site,TATAbox,CAATbox,in the 1705bp promoter region. The binary vector harboring AtCKX1 driven by RD29B promoter was further constructed to be used in coming plant transgene.
